City beat Feyenoord and Shakhtar edged past Napoli in gameweek 1 of this season's Champions League.
Pep Guardiola's side will be hoping to build on the strong start, but will come up against stern opposition in the Ukrainian side.
Manchester City:
City will be without captain Vincent Kompany and Ilkay Gundogan, as both have failed to overcome short term injuries in time to feature.
Full-back Benjamin Mendy will undergo a late fitness test, while Danilo will be on hand to deputise if the former Monaco man is deemed not ready to play.
Guardiola will most likely start Aguero and Jesus up front and hope they can continue their prolific form in front of goal.
Shakhtar Donetsk:
The visitors will be without Darijo Srna, who recently tested positive for a banned substance and stated he would stay away from football until he clears his name.
Maksym Malyshev and Serhiy Kryvtsov are the other long-term absentees.
